Ryan Gleason of the Padgett and Cai labs will present his research. Below is a short description he has provided.
To identify components of the TGFb superfamily signaling pathways the Padgett Lab has conducted both a F2 clonal genetic screen and a suppressor screen to isolate mutants in the nematode, Caenorhabditis elegans. Further EMS mutagenesis screens have provided phenotypically tractable models that have established conserved members of the canonical pathway and additional regulatory members. Two alleles, wk94 and Sma-10, identified in this screen are poised to give new insight into signal transduction and regulation of the pathway.
With conserved structural features and sequence homology TGFb ligands have been identified throughout metazoan biology. The TGFb superfamily is capable of regulating a compelling array of cellular processes, including both stimulatory and inhibitory regulation of cell proliferation, cell migration, cell death, immune regulation, embryonic stem cell self-renewal, and differentiation of somatic stem cells. How does one pathway achieve such diverse and at times opposing biological roles? The result of TGFb signal transduction is dependent on the cell type and cell context in which the signal is received. Various sites of regulation such as the extracellular milieu, endocytic trafficking, protein expression, and transcriptional networks are exploited to fine-tune the pathway to elicit the precise response. Studies to delineate the regulation and function of Sma-10 and wk94 suggest contribution of these members at the various sites of regulation above.
Cell signaling cascades that regulate stem cell self-renewal and differentiation have been intensely studied. The TGFb signaling cascade, along with other signaling cascades, maintain the self-renewal and differentiation of both embryonic stem cells and somatic stem cells. Intriguingly, TGFb superfamily ligands have disparate roles in regulating the biology of embryonic stem cells compared with neural stem cells. In collaboration with the Cai Lab, investigation is underway to identify and delineate the contribution of these new members and their role in neural differentiation.

Our IRIF presenter this month is Andrea Gray of the Yarmush and Banerjee labs. She will be presenting her research focusing on computational modeling and magnetic targeting of encapsulated MSCs for spinal cord injury. The computational model is a three-dimensional representation of the human spinal subarachnoid space in which computational fluid dynamics will be used to study the transport of secreted factors (e.g. cytokines) through the cerebrospinal fluid from the encapsulated MSCs to the injury site and vice versa. In parallel, magnetic nanoparticles will be co-encapsulated with the MSCs to facilitate positioning within and potential removal of the capsules from the spinal subarachnoid space using an external magnetic field. The goals of these approaches are to determine if lumbar placement of encapsulated MSCs is sufficient for injuries of variable location and to enable targeted delivery of the capsules; achievement of these goals will result in a more controllable cellular therapy for spinal cord injury. |

Ian Gaudet of the Shreiber, Firestein and Burdick labs will be presenting his research focusing on the synthesis, characterization, and application of a novel collagen-PEG hydrogel. This hybrid material is photo-crosslinkable and self assembling, which allows for patterning of heterogenous mechanical properties within a 3D scaffold in the presence of live cells. The eventual goal is to be able to spatially direct hMSC differentiation into neural lineages via manipulation of the substrate compliance. |

Research involves the differentiation of embryonic stem cells into pancreatic endocrine cells, such as insulin producing beta cells. These cells can later be transplanted into Type 1 diabetes mellitus patients to help maintain glucose levels within a healthy range. In addition, he is exploring the ability of the differentiated cells to vascularize in the recipient’s tissue to improve cell’s engraftment, survival and function. |
